I doubt the quill lock will affect this much. All it does is prevent the quill feed shaft from turning. It might be slightly less, but I do not think it will be much.
Regardless, run out(slop) is a negative factor as the quill is extended and retracted(drilling). Spindle routing may be affected(quill stationary), but we be splitting hairs there!
Maybe even this whole(from post 8 on) thread is splitting hairs!;)

In the absence of of a dial gauge, I fitted the Jacobs chuck with a 1/8" 60 degree V bit and clamped a piece of ply to the table so that with the quill a quarter inch short of full extension the sharp of the V bit touched the edge of the ply.
With the quill unlocked I then rocked the quill (one hand holding the quill lever) from side to side, making the longest line I could in the clamped up ply.
