What’s the best way to cut a notch out of plexiglass. My daughter wants me to cut out a notch on a piece of plexiglass.

If you have one, I would use a jigsaw or a scrollsaw.
If blade of whatever you are cutting with gets too hot the plexiglass will melt and then reform behind the blade.

I have scroll saw and coping saw. One of my problems I have no scrap to practice on so I have to be careful and use best option.

Since you do not have any scrap to practice on I would go with the coping saw.

I've cut Lexan with the TS, BS, Scroll saw and a jig saw. I leave the paper on, though. It helps with the melting you get with the slower saws. The scroller, especially.

Thanks everyone, I taped both sides and used metal cutting blade on band saw and it came out fine. Once again, thanks!

Sorry I missed this discussion. Was going to say I have had success with the band saw.

I have cut a lot of plexi just with the table saw. It cuts it well, although you want to watch for vibration. A heavier piece of wood along the side of the piece you are saving is a good idea, just to keep it flat. I just used my regular Forrest Woodworker II blade.
